Based on the Gillian Fynn's best-selling mystery, the 21-year-old model will play Andie, the young mistress of the married Nick Dunne, played by Affleck.
When Nick's wife goes missing, Andie joins the majority of characters who cast a suspicious eye on the unhappily married man, believing foul play might be involved.
This is Ratajkowski's first major screen role. Also starring in Gone Girl is Rosamund Pike, Neil Patrick Harris, Tyler Perry and Patrick Fugit.
The model who was previously seen in ads for Forever 21 and Carl's Jr. first made waves on Nickelodeon's iCarly as Tasha.
Earlier this year, the stunner nabbed national attention when she appeared topless in Robin Thicke's Blurred Lines video.
Ratajkowski will next appear in an upcoming episode of New Girl.
